Pharmacology

TAK-071 is a novel, potent and highly selective muscarinic acetylcholine receptor 1 (M1R) positive allosteric modulator. It lacks agonist affinity compared to other M1 modulators, improving efficacy and tolerability. It is similar to VU0453595 and VU0550164 (other M1 PAMs), but is likely superior, and also has the benefit of having human trials.

Overall, TAK‐071 was safe and well tolerated at the doses tested. There were no serious AEs or deaths.

Estimated dose is 1mg IN. Half life was found in the first safety study to be 45-60 hours, however it may be around half for lower dosages than what they used.
