Neboglamine
C+

Pharmacology

Neboglamine is a small molecule NMDA Glycine PAM that is considered a nootropic. Efficacy in healthy models is limited, and it has potential downsides.

The brain naturally produces a neurotransmitter named D-Serine, and Neboglamine potentiates its binding co-agonist site, specifically. Saying that, it has some noticable differences.

Neboglamine is similar to D-Serine, but lacks the NR3 selectivity leading to potential dentritic shortening, which would be opposite of NR2 enhancement via the Glycine site. The NR3 site only requires glycine (NR1 binds to both NR2 and NR3 but only requires glycine too), while NR2 requires glutamate, meaning the modulation of NR3 by Neboglamine may be more due to this.

Neboglamine is a relatively effective antidepressant, but the short half life is not ideal.

Generally, something potentiating a more selective subtype (such as NR2B like Rapastinel derivatives), or something potentiating endogenous D-Serine (such as Luvadaxistat), or other NMDA modulators in general are likely better.

Neboglamine also has poor pharmacokinetics, with poor solubility outside of the HCl version, and an approx dose of 50-75mg in humans. It's half life is also about 4 hours approximately. It is also a weak norepinephrine uptake inhibitor (NRI), and is more apparent at dosages over 100mg.
